1st time? You only fight Python once. I think you mean Null the first time. So I'll give you both.
Python.
Forget all the other strategies for one, including the crap of running to the second floor. You can beat him with relative ease by following my simple strategy:
Needed Items: You only get 4, so you have to be carefull. Make sure to have Body Armor, Medica Kits (Large preferably), Mosin Nagant. The forth item is the C4 you use to finish the mission so it's there but you won't need it for the actual fight.
In the room you start out in there are two elevated metal "thing" in the top and bottom corners of the room. So when the fight starts run forward, right past Python. Head to the right top corner of the room. The elevated platform is like waist high so just climp on it and get to the wall. Lay down facing an opening. Now you have a perfect view of Python as he can only come right infront of you to get take his shot. The metal box thing will protect you from all other sides.
Equip the body armor and get your Mosin Nagant ready. Don't waist time with blind firing, make your shots count. You only need 3 headshots to take him out. That's all. The fight is very quick if you just aim for his head. Plus it's a tranq, so he will join your team right after the fight.
Null (1st fight) - easiest boss to beat of all!!!
Items needed - Any handgun will do, even an MK22.
As the fight starts you are near a building where the goal of the mission was. To your left there is a crawl space under the building. Go in there. Keep crawling until you crawl out on the other side. Keep crawling. Right infront of you will be these metal contaners you can crawl under. While you are under the building Null will hide from your view. He will do the same as long as you are under the container.
As you are under the container, face the corner where Null is (it should be the one right next the building crawl space you came out of). Crawl backwords, just slightly. Null will come out and basically begin walking infront of the container, side to side, witout ever goind around the darn thing to get to you!!!! Just shoot his legs!!! He will keep returning to the same spot. The most retarded fight EVER! Just lay there and shoot his legs as he walks from side to side. It's a glitch in the AI, but it works to your advantage.

That all depend on what you are trying to do. You can run through the game fairly fast if you don't bother recruiting. Each mission is fairly small. However, you'll be barely making it since you will be low on supplies and intel.
The first time I went through it too 21 hours total. (MGS3 took me 22 hours the first time). The second run only lasted 11 hours. Now the thing of it is, I recurited a lot during both.
If you just do the core game without side missions and minimal recruiting you can probably run through it in a few hours. Can't tell you for sure yet. Once I'm happy with the unit and all the characters are unlocked, I'll run through it just to run through it for the time.
